A karyotypic study of three southern Brazilian Asteraceae species using fluorescence in situ hybridization with a 45S rDNA probe and C-CMA3 banding Genet. Mol. Biol.
Fregonezi,Jéferson N.; Torezan,José M.D.; Vanzela,André L.L..
The Asteraceae, one of the largest families of flowering plants, contains about 1,100 genera and 20,000 species, and is well known for its extensive karyotypic variation. In this study, conventional Feulgen staining, C-CMA3 banding, and fluorescence in situ hybridization with a 45S rDNA probe were used to determine the chromosome number and the number and physical position of GC-rich heterochromatin and 45S rDNA sites in three Asteraceae weed species (Crepis japonica, Galinsoga parviflora and Chaptalia nutans). The three species exhibited karyotype differences in the chromosome number and shape, as a commom feature of Asteraceae. However, the 45S rDNA sites always occurred on the short chromosomal arms, associated with GC-heterochromatin. Althought of...

Chromosome mapping of 5S rRNA genes differentiates Brazilian populations of Leporellus vittatus (Anostomidae, Characiformes) Genet. Mol. Biol.
Aguilar,Cecilia Teixeira de; Galetti Junior,Pedro Manoel.
Among the anostomid fishes, the genus Leporellus is represented by only three species: L. nattereri, endemic of the Amazon River, L. retropinnis, endemic of the Piracicaba River, and L. vittatus, widely distributed in rivers from Peru, Colombia, Guianas, and different major hydrographic basins of Brazil. A cytogenetic study carried out on specimens of Leporellus vittatus from three major Brazilian hydrographic basins evidenced a karyotype of 54 metacentric and submetacentric chromosomes. C-banding analysis revealed the presence of large pericentromeric heterochromatic segments in all chromosomes and a telomeric block coincident with the NOR sites. Ag, CMA3 or MM staining, and FISH with ribosomal probes located the 45S ribosomal genes on the terminal region...

Localization of 45S rDNA and telomeric sites on holocentric chromosomes of Rhynchospora tenuis Link (Cyperaceae) Genet. Mol. Biol.
Vanzela,André L.L.; Cuadrado,Angeles; Guerra,Marcelo.
Rhynchospora tenuis Link (Cyperaceae) is a weed widely distributed in Brazil that presents a small number of holocentric chromosomes (2n = 4) with some autopolyploid populations (2n = 8). The haploid number n = 2 is considered as a derivative of the base number x = 5. 45S rDNA probes and telomeric DNA were hybridized in both chromosome races of R. tenuis, looking for indications of chromosome fusions. The results showed that hybridization sites of the telomeric probe were restricted to end chromosome regions whereas rDNA sites were terminally located. The chromosome race with n = 4 exhibited a doubled number of sites, with similar size and location to the hybridized sequences, confirming its autopolyploid origin. Furthermore, the terminal location of the...

Morphological characterization under different ecological habitats and physical mapping of 5S and 45S rDNA in Lilium distichum with fluorescence in situ hybridization RChHN
Hwang,Yoon-Jung; Song,Chang-Min; Younis,Adnan; Kim,Chang-Kil; Kang,Yoon-Im; Lim,Ki-Byung.
BACKGROUND: This study was performed to investigate the phenotypic, karyomorphological, and habitat environment characteristics of Lilium distichum that grows naturally in South Korea. Currently, this species follows limited distribution areas and its natural populations are at the brink of extinction mainly due to fragmentation or destruction its natural habitat. RESULTS: This species was distributed between approximately 1,000 and 1,500 m above sea level with an average temperature of 22°C. The soil characteristics surrounding the natural habitats included loamy and silt loam soils having organic matter content (10.82%), pH (5.22), electrical conductivity (EC) (0.37 dS/m), total nitrogen (0.45%), and cation exchange capacity (34.3 cmol+/kg). The peak...
